James carried out his dissertation research in Dr. Joe Loparo's lab, studying how multiple types of DNA polymerases coordinate and exchange to replicate at sites of mutagenic DNA damage in Escherichia coli. For this project, he developed novel single-molecule approaches, as well as using the standard techniques of protein biochemistry and bacterial genetics.
